The COT One Step Cotinine Test Device (Urine) is a lateral flow chromatographic immunoassay for the detection of Cotinine in human urine at a cut-off concentration of 200 ng/mL. Cotinine is a principle and residual metabolite of Nicotine which remains in the body of habitual tobacco users.

This test is intended for forensic, non-diagnostic use only to determine the smoking status and use of tobacco/nicotine only. This test is not intended to diagnose any medical condition or aliment. It is used for forensic use only.

The COT One Step Cotinine Test Device (Urine) provides only a qualitative, preliminary analytical result. A secondary analytical method must be used to obtain a confirmed result. Gas chromatography/mass spectrometry (GC/MS) is the preferred confirmatory method.1

• Allow Test Device & Urine Specimen to reach room temperature (15-300C) prior to testing.
• 1. When ready to perform the test, remove the test device from the pouch. Discard the small moisture absorbent packet.
• 2. Place the test device on a clean and level surface. Hold the dropper vertically and transfer 3 full drops of urine to the specimen well (S) of the test device and start the timer for 5 minutes. Avoid trapping bubbles in the specimen well (S).
• 3. Wait for the colored lines to appear. The results should be read at 5 minutes. It is important that the background is clear before the result is read. Do not interpret the results after 10 minutes.

• POSITIVE RESULT: One red line appears in the Control (C) region. No line appears in the Test (T) region.
• NEGATIVE RESULT: Two red lines appear. One red line should be in the Control (C) region & another red or pink line should be in the Test (T) region.
• Note: The shade of color in the test region (T) may vary, but it should be considered negative even when there is even a faint line.
• INVALID RESULT: Control (C) line fails to appear. Insufficient specimen volume or Incorrect procedural techniques are the most likely reasons for the Control line failure.
• LIMITATIONS:
• 1. The One Step test Strip provides only a qualitative, preliminary analytical result. A secondary analytical method must be used to obtain a confirmed result. Gas Chromatography/Mass Spectrometry (GC/MS) are the preferred confirmatory methods.
• 2. It is possible that technical or procedural errors, as well as other interfering substances in the urine specimen may cause erroneous results.
• 3. A Positive Result indicates presence of the drug or its metabolites but does not indicate level of intoxication, route of administration or concentration in urine.
• 4. A Negative Result may not necessarily indicate drug-free urine. Negative Results may be obtained when drug is present but below the cutoff level of the test.
